KRAS G12V, BRAFV600E and HRASG12V oncogenic effects on the ability of colon adenocarcinoma cells to migrate and invade in vitro. (A) Cells were seeded on a polycarbonate membrane and allowed to migrate for 30 hours. Cells that migrated through the surface of the membrane were stained with crystal violet and counted by bright field microscopy. (B) For the invasion assay cell were seed on polycarbonate membrane coated with matrigel and incubate for 36 hours. (C) HT29 cells transfected with shRNA-BRAFV600E (HTshB3) or empty vector (HTps) and DLD-1 cells before and after knockout of KRASG13D (DKO4). Migration assay was performed as mentioned in (A). *: P ≤ 0,05; **: P ≤ 0,01; ***: P ≤ 0,001 as determined with Student t-test.
