HBx-Tg mice exhibit an increased oxidative stress and susceptibility to liver ischemia-reperfusion challenge. (A) The transcript of HBx in livers from HBx-Tg or wide type (WT) mice was detected using RT-PCR. (B) Mice were given butylated hydroxyanisole (BHA)-containing (0.7%) or control diet for 2 days and then subjected to 60 min of warm liver ischemia (see "Materials and Methods"), followed by 6 hr of reperfusion before sacrificed. Total liver GSH content and GSH/GSSG ratio were determined. Values are mean ± SD (n = 4). *, p < 0.05. (C) Frozen liver sections from mice treated as in Fig. 1B were subjected to dihydroethidine (DHE) staining. Representative results are shown. Magnification, ×100. (D) Livers from mice treated as in Fig. 1B were homogenized and the protein levels of PARP and GAPDH were determined by Western blot assay. GAPDH was used as a loading control.
