Figure Enforced expression of ectopic Mcl-1 in A549 cells protects cells from apoptosis induced by the API-1 and TRAIL combination. A, The indicated stable transfectants seeded in 96 well-plates were treated with 5 μM API-1 alone, the indicated concentrations of TRAIL alone and API-1 in combination with TRAIL. After 24 h, the cell numbers were estimated with SRB assay. The data are means ± SDs of four replicate determinations. B, The indicated stable transfectants were treated with 5 μM API-1 alone, 30 ng/ml TRAIL alone or their combination for 22 h and then harvested for preparation of whole-cell protein lysates and subsequent Western blot analysis to detect PARP cleavage.
