An Akt/ERK-miR-200c-ZEB2 axis was involved in IGF-induced EMT in gastric cancer cells. MGC-803 and SGC-7901 cells were serum-starved overnight and then treated with or without 100 ng/mL of IGF-I for 48 h. (A) The expression of ZEB2 was analyzed by Western blot. (B) The level of mRNA for ZEB2 was analyzed by RT-PCR. (C-E) The serum-starved cells were pretreated with or without PD98059 (20 μM) or LY294002 (100 μM) for 2 h followed by IGF-I (100 ng/mL) stimulation for 48 h. The expression of miR-200c was analyzed by real-time PCR. Data are means ± SD in three independent experiments. * IGF-I untreated vs. IGF-I treated, p < 0.05. Control group as reference.
