Effects of AGR2 knockdown on TPC-1 cell growth, migration and invasion. A) Expression levels of AGR2 in Nthy-ori 3–1 and TPC-1 cells. Cells were lysed and analyzed by Western blotting with the indicated antibodies. B) TPC-1 cells were transfected with AGR2 siRNA or with scrambled siRNA and counted at different time points. Values represent the average of triplicate experiments ± standard deviations. C) TPC-1 cells were transfected with AGR2 siRNA or with scrambled siRNA; after 48 and 72 hours cells were collected by trypsinization, stained for 10 minutes with trypan-blue and counted in triplicate. The percentage of trypan blue excluding cells compared to cells transfected with scrambled siRNA is reported ± standard deviations. D) TPC-1 cells were transfected with AGR2 siRNA or with scrambled siRNA; after 48 hours a wound was introduced and cell migration into the wound was monitored at 12 and 24 hours.
