SPARC boosts in vivo growth and metastatic dissemination of M cells. (a) SCID-Beige mice (n =10 / experimental group) were inoculated orthotopically (dorsal prostate) with 1x105 firefly luciferase-expressing M.Fluc and S.Fluc.sh3399 cells, as indicated, and growth monitored by external BLI. Mice implanted with S.Fluc.sh3399 cells integrating doxycycline-inducible SPARC-specific shRNA were administered, or not, doxycycline for the entire duration of monitoring (+dox or -dox, respectively). (b) Representative images corresponding to mice in the same experimental groups as in (a). (c, d) Upon termination of the in vivo bioluminescent monitoring (day 24), prostates (c) and lungs (d) from inoculated animals were removed, incubated with D-luciferin and photon counts determined. Asterisks denote statistically significant differences (two-tailed Mann–Whitney test). Dotted lines represent non-linear fitting to the exponential growth equation Y = Y0(kX).
