From: p21WAF1 expression induced by MEK/ERK pathway activation or inhibition correlates with growth arrest, myogenic differentiation and onco-phenotype reversal in rhabdomyosarcoma cells

Figure 9

Enforced p21WAF1 expression induces growth arrest in RD cells. (A) Immunoblotting using specific antibody capable of recognising the p21WAF1 of total lysates from RD cells transfected with CB6/p21 or the empty vector (CB6), cultured without (-) or with 120 μM ZnCl2 (+) for 3 days before (Transient transfection) and after (Stable transfection) neomycin-selection. (B) Growth graph of same neomycin-selected RD cells cultured as in A. As a control, untransfected RD cells were left untreated (C) or treated with TPA or U0126. (C) Cell cycle distribution of RD cells transfected with empty vector PEGFP (left) or with PEGFP/p21 (right). Cell cycle distribution of GFP-expressing cells was evaluated by flow cytometry. The data shown in the insert tables are the mean ± s.e.m. of triplicates of a representative experiment. Similar results were obtained in two experiments.
