PTEN knockdown does not alter proteasome-dependent degradation. Cells stably transfected with PTEN-shRNA or empty vector were treated with three different proteasome inhibitors (lactastatin, 20 mM; LLnL, 50 mM; MG132, 1 mM) or cycloheximide (CHX; 20 μg/ml) for 6 h, followed by immunoblotting for p21.
