High concentrations of nilotinib down-regulate the expression of FoxP3 and GITR by CD4⁺CD25⁺ T cells. Purified CD4⁺CD25⁺ T cells were unstimulated or stimulated with anti-CD3, anti-CD28 and IL-2 in the presence or absence of nilotinib as indicated. Then cells were stained with FoxP3*PE or GITR*PE according to the manufacturer's instruction for fixation, permeabilization, and staining, after cells were stained for surface expression of CD4 and CD25. Nilotinib significantly decreased the intracellular expression of FoxP3 and GITR on CD4⁺CD25⁺ T cells in a dose-dependent manner at concentrations of 25 μM.
