DIF-1 inhibited HUVECs proliferation. (A) Cell proliferation assay. HUVECs seeded on a 24-well plate (1.0×104 cells/well) were incubated with various amount of DIF-1. Cells were harvested by trypsin/EDTA treatment at the times indicated and enumerated. Values are mean ± SE of three independent experiments and statistically analyzed using a one-way ANOVA with Bonferroni post-hoc test. The asterisk indicates *P < 0.05 and **P < 0.01 versus control. (B) Flow cytometry. HUVECs were incubated with DIF-1 (30 μM) for 24 h and then harvested by the trypsin/EDTA treatment. Cells were stained with propidium iodide (PI) and fluorescence of nuclei was measured. The percentages of cell number in the cell cycle phases are also shown. The results are means ± SE of three independent experiments. The asterisk indicates *P < 0.01 and **P < 0.001 versus control.
