Activation of AP-1 by LPA, EGF or HGF. A. Caov-3 cells were starved and stimulated for the indicated periods of time with LPA (10 μM), EGF (25 ng/ml) or HGF (20 ng/ml). The cell lysates were analyzed by immunoblotting for expression of AP-1 proteins c-Jun, c-Fos, Jun-B and Fra-1 and other loading controls (lamin A/C, β actin or tubulin). B. Caov-3 cells were stimulated as in A and analyzed for AP-1 DNA binding activity with EMSA by incubating with 32P-labeled AP-1 consensus oligonucleotides. Reaction mixes were run on 5% native polyacrylamide gels and autoradiographed. The specificity of binding to the 32P-labeled AP-1 probe in nuclear extracts of cells treated with LPA for 2 hours was confirmed by inhibition of the binding with 50 times of unlabeled oligonucleotides (cold competition).
