ADAM12 knockdown reduces tumor formation in NOD-SCID mice after orthotopic cell transplantation. a Tumor incidence in the limiting dilution transplantation assay. SUM159PT_shADAM12 and SUM159PT_shControl cells were transplanted in the fourth mammary fat pad of NOD-SCID mice in limiting dilutions (10,000, 1,000, or 100 cells per injection site). Prior to transplantation, cells were pre-treated with doxycycline (Dox; 1 μg/ml) for 4 days, and doxycycline (2 g/kg) was then continually administered in the diet. The presence of palpable tumors was determined 3 weeks after cell transplantation. The frequency of breast tumor initiating cells (BTICs) and P-values for the statistical significance between groups were calculated using extreme limiting dilution analysis (ELDA, http://bioinf.wehi.edu.au/software/elda/; ref.
