From: Long non-coding RNA PVT1 promotes tumor progression by regulating the miR-143/HK2 axis in gallbladder cancer

Fig. 3

PVT1 negatively regulates miR-143 expression. (a) The potential binding sites between miR-143 and PVT1. (b-c) miR-143 was downregulated in GBC tissues, and PVT1 expression and miR-143 expression were inversely correlated in GBC tissues. (d) MiR-143 expression was increased after PVT1 knockdown. (e-f) GBC-SD and NOZ cells were transfected with pcDNA-NC or pcDNA-PVT1. qPCR showed that pcDNA-PVT1 upregulated PVT1 expression and decreased miR-143 expression. (g-h) The expression level of PVT1 was decreased after transfection with miR-143 mimics but increased after transfection with miR-143 inhibitor. (i) Co-transfection of miR-143 mimics and pmirGLO-PVT1-wt strongly decreased the luciferase activity, while co-transfection of miR-143 mimics and pmirGLO-PVT1-mut did not change the luciferase activity. *P < 0.05, **P < 0.01, ***P < 0.001.
