Effect of B7.1 silencing on the Ad5FB4-DA1-3b/d365 cell binding. Cells were electroporated with control siRNA or siRNA against B7.1 (3 nmol/cell), maintained in culture for 96 h, transferred to 4°C, then incubated with FAM-labeled Ad5FB4 at 104 vp/cell for 90 min at 4°C. (A), The efficiency of B7.1 silencing was verified by flow cytometry, using PE-labeled antibody against murine B7.1. (a), control siRNA; (b), siRNA against B7.1; (c), control irrelevant isotypic antibody. (B), Cell-bound Ad5FB4 vector particles were quantitated by flow cytometry analysis of cell surface-associated fluorescent signal, and results expressed as the percentage of positive, fluorescent cells.
