FKB induced G 2 /M arrest in OS cells. A, 143B and Saos-2 cells were treated with vehicle control or FKB at different concentrations for 24 h and analyzed for cell cycle. Percentage of cell cycle distribution after FKB treatment was presented. Points, mean of three samples for each concentration; bars, SE. B, 143B cells were blocked at G2/M phase and released form the block into either control or 5 μg/ml FKB. At times indicated, cells were collected, fixed, and analyzed by flow cytometry. C, The effect of FKB (5 μg/ml) on cell-cycle regulatory markers was examined by Western blot analysis. The protein extracted from 143B cells treated with FKB for indicated time was resolved by SDS-PAGE.
