Influence of MLH1 knock down by siRNA treatment. Direct connection of MLH1 on SPTAN1 was studied in detail by comparison of untreated with transiently MLH1-specific siRNA1 (A) and siRNA2 (B) and control siRNA transfected different MLH1 proficient cell lines. Western blot analysis of MLH1 and SPTAN1 protein levels was performed after 48 h, controlled by beta-Actin detection after. In parallel, mRNA levels of MLH1 and SPTAN1 were determined by TaqMan-analysis with pairs of MLH1-, SPTAN1- or GAPDH-specific primers and MLH1-, SPTAN1- or GAPDH-specific probes, respectively, after treatment with MLH1-specific siRNA1 (C) and siRNA2 (D). The data show that siRNA knock down of MLH1 led to impaired SPTAN1 expression and partly to reduced mRNA level of SPTAN1.
