Location of endogenous parafibromin in HEK293 cells transfected with actinin. HEK293 cells transfected with plasmid expressing FLAG-actinin-2 were analyzed by immunofluorescence with primary antibodies that recognize parafibromin (GRAPE) or the FLAG-tag (actinin-2), and secondary antibody conjugated to Texas Red (B, to detect endogenous parafibromin) or fluorescein isothiocyanate (C, to detect transfected actinin-2), followed by DAPI staining (A). Note that in untransfected cells parafibromin is nuclear but in the 3 cells over-expressing FLAG-actinin-2, parafibromin is in the cytoplasm.
