ChIP assay. Esophageal cancer TE-3 cells were grown and treated with 1 μM BPDE for up to 24 h. After that, the cells were fixed with formaldehyde for 10 min, and the nuclei were then isolated, sonicated, and immunoprecipitated with anti-BPDE antibodies. (A) Immunoprecipitated protein was subjected to Western blotting analysis of DNMT3A and DNMT3B expression. Protein without anti-BPDE antibody immunoprecipitation (input) was used as the control. PC, positive control. (B) Immunoprecipitated DNA was subjected to PCR analysis of RAR-β2 gene promoter.
