Effect of L-Asparaginase on SP cells. a Conversion of asparagine to asparatic acid or glutamine to glutamate catalyzed by asparaginase. b Generation of glutamate from glutamine by L-Asparaginase. Cell-free medium containing glutamine (30 mg/dl) was incubated with the indicated concentrations of L-Asparaginase for 5 h, and medium was collected to for measurement of glutamate. c Cell-free medium containing glutamine (30 mg/dl) was incubated with 1U/ml L-Asparaginase for indicated time, and the medium was collected for detection of glutamate. d A549 Cells were incubated with 1U/ml L-Asparaginase (L-ASP) for the indicated times. Then cells were harvested and stained with Hoechst 33342 to determine SP fraction. The number (%) within each panel indicate the percentage of SP cells in the whole cell population
