The role of BRD4 isoforms in DNA damage response. BRD4-S stably binds to lysine-acetylated histones (Ac), promoting chromatin compaction and shielding damaged DNA from the DNA damage repair (DDR) machinery. BRD4-L promotes non-homologous end joining (NHEJ) DNA repair. BRD4-L induces NHEJ genes transcription and stabilizes NHEJ protein complexes in the DNA damage site. Increased activity of either or both isoforms can lead to genomic instability due to decreased DDR and promotion of error-prone NHEJ.
